DMEM/F-12 (Dulbecco’s Modified Eagle Medium/Nutrient Mixture F-12) is a widely used basal medium that can be used to support the growth of many types of mammalian cells. Cells successfully cultured in DMEM/F-12 include MDCK, glial cells, fibroblasts, human endothelial cells, and rat fibroblasts.
DMEM/F12 medium is based on DMEM medium, adding richer nutrients in F12 medium, containing a variety of trace elements, and is widely used in the cultivation of various mammalian cells. At the same time, DMEM/F12 medium is often used as the basis for developing serum-free medium, and it is also suitable for the culture of mammalian cells and colony density culture under low serum content.DMEM/F-12 is a 1:1 mixture of DMEM and Ham’s F-12, containing DMEM’s high concentration of glucose, amino acids and vitamins with various components of F-12. DMEM/F-12 does not contain proteins, lipids or growth factors. Therefore, DMEM/F-12 may require nutritional supplementation, usually with 10% fetal bovine serum (FBS). DMEM/F-12 uses a sodium bicarbonate buffer system and therefore requires a 5–10% CO2 environment to maintain physiological pH.

This DMEM/F-12 dry powder cell culture medium needs to be supplemented with sodium bicarbonate, adjusted for pH and filtered during preparation. The detailed formulation is as follows:
Add 950 mL of distilled water to a mixing vessel as close to final volume as possible.
Add dry powdered medium to room temperature (15°C to 30°C) water and stir gently. Do not heat water.
Rinse the inside of the pack to remove any residual powder.
Add 50mL of 75g/L NaHCO3 solution to the culture medium.
Adjust the pH to 0.2 to 0.3 units below the desired final working pH by slowly adding and stirring 1 N NaOH or 1 N HCl. The pH may rise by 0.1 to 0.3 units after filtration. The recommended working pH is 7.0-7.4.
Use distilled water to adjust the final volume.
Immediately process the culture medium through a 0.2 µm filter membrane into sterile containers using a positive pressure filtration system.
