Powdered Leibovitz L-15 Medium

L-15 is buffered with phosphate and free amino acids instead of sodium bicarbonate. This medium is suitable for supporting cell growth in a non-CO2 balanced environment. L-15 contains no proteins, lipids, or growth factors. Therefore, F-10 requires additives, typically with 10% fetal bovine serum (FBS).

Ingredients of Leibovitz L-15 Medium

This Leibovitz L-15 dry powder medium is formulated with pH adjustment and filtration. The detailed formulation scheme is as follows:

1. Add 950 mL of distilled water to the mixing vessel as close as possible to the final volume.

2. Add dry powder medium to room temperature (15 °C to 30 °C) water and stir gently. Do not heat the water.

3. Rinse the inside of the package to remove any residual powder.

Adjust the pH to 7.6 by slowly adding 1 N NaOH or 1 N HCl and stirring.

5. Use distilled water to adjust the final volume.

6. Immediately use a positive pressure filtration system to process the medium into a sterile container through a 0.2 μm filtration membrane.
